ABSTRACT

Abstract The present study aimed to investigate the beneficial of prepared black rice anthocyanins nano-composite (An-AgNps) against hepatotoxicity induced by methotrexate (MTX) in rats. Anthocyanins nano-composite was prepared by silver as the metallic ion reduction and were characterized by IR and SEM. The rats in our experiment were divided into five groups. Serum lipid profile, serum transaminases (ALT and AST), ALP, LDH, TBA, GSH and SOD were examined. The results show that SEM of An-AgNps has average particle size from 70 to 130nm. In the group treated with MTX; TC, TG, LDL-c, ALT, AST, ALP, LDH and TBA levels were significantly (P0.05) increased than NC, while, HDL-c, SOD and GSH levels were significantly (P0.05) decreased. On the other hand, An-AgNps + MTX treated groups were reversed the levels of all biomarkers similar to NC. In conclusion, the results show that An-AgNps has a protective effect on MTX-induced hepatotoxicity and oxidative stress.


Resumo O presente estudo teve como objetivo investigar o benefício de nanocompósito de antocianinas de arroz preto preparado (An-AgNps) contra a hepatotoxicidade induzida por metotrexato (MTX) em ratos. O nanocompósito de antocianinas foi preparado a partir da prata por meio da redução do íon metálico e caracterizado por IR e SEM. Os ratos em nosso experimento foram divididos em cinco grupos, e foram examinados o perfil lipídico sérico, as transaminases séricas (ALT e AST), ALP, LDH, TBA, GSH e SOD. Os resultados mostram que SEM de An-AgNps tem tamanho médio de partícula de 70 a 130 nm. No grupo tratado com MTX, os níveis de TC, TG, LDL-c, ALT, AST, ALP, LDH e TBA aumentaram significativamente (P 0,05) do que NC, enquanto os níveis de HDL-c, SOD e GSH diminuíram significativamente (P 0,05). Por outro lado, nos grupos tratados com An-AgNps + MTX, foram revertidos os níveis de todos os biomarcadores semelhantes ao NC. Em conclusão, os resultados mostram que o An-AgNps tem um efeito protetor contra a hepatotoxicidade induzida pelo MTX e o estresse oxidativo.

ABSTRACT

Background: Berberis commutata Eichler is a berry that grows in the Peruvian Andes and has been consumed in the Andes of South America since ancient times. The edible fruits have an intense purple color and are rich in anthocyanins and phenolic compounds that are available from February until May each year. The color of the fruits is a soft purple dye for natural fibers, and many birds use them as food. Objective: This study quantified the total phenolic, monomeric anthocyanin content and antioxidant activity of Berberis commutata Eichler berries. Methods: The total phenolic content was determined by the Folin-Ciocalteu colorimetric assay. Monomeric anthocyanin content was determined by the method is pH differential, and the antioxidant activity was measured using the Brand-Williams method. Results: The total phenolic content was 7,490 ± 0.85 mg GAE/100g, and the monomeric anthocyanin content was 70 ± 0.03 mg/100g. The antioxidant activity of the berries showed a tendency to increase with B. commutata extract concentration; an EC50 of 0.91 mg/mL was calculated, indicating a high antioxidant power. Conclusion: Our results showed that B. commutata E. has both high total phenolic content and monomeric anthocyanins comparable to other superfruits and high antioxidant activity, which means that it is possible to use this berberis species as a functional food.


Introducción: Berberis commutata Eichleres una baya que crece en los Andes peruanos. Los frutos comestibles tienen un intenso color púrpura rico en antocianinas y componentes fenólicos que están disponibles desde febrero hasta mayo de cada año. El color de sus frutos se utiliza como un suave colorante púrpura para las fibras naturales y muchas aves los utilizan como alimento. Sin embargo, desde la antigüedad los frutos de esta especie han sido consumidas en los Andes de Sudamérica. Objetivo: Este estudio cuantificó el contenido fenólico total, antocianinas monoméricas y la actividad antioxidante usando el método del radical DPPH de las bayas de Berberis commutata Eichler. Método: El contenido fenólico total se midió a través del ensayo colorimétrico de Folin-Ciocalteu, el contenido de antocianinas monoméricas se determinó mediante el método del pH diferencial y la actividad antioxidante se midió con el método de Brand-Williams. Resultados: El contenido fenólico total fue de 7,490 ± 0.85 mg GAE/100g y el contenido de antocianinas monoméricas fue de 70 ± 0.03 mg/100g. La actividad antioxidante de las bayas mostró una tendencia a aumentar con la concentración del extracto de B. commutata, se calculó un EC50 de 0.91 mg/mL que indica un alto poder antioxidante. Conclusión: Nuestros resultados mostraron que B. commutata E. tiene tanto un alto contenido fenólico total, así como antocianinas monoméricas comparables con otras superfrutas y una elevada actividad antioxidante, lo que significa que es posible utilizar esta especie de berberis como alimento funcional.

ABSTRACT

Studies have shown that targeting xanthine oxidase (XO) can be a feasible treatment for fructose-induced hyperuricemia and hyperglycemia. This study aimed to evaluate the dual regulatory effects and molecular mechanisms of diacylated anthocyanins from purple sweet potato (diacylated AF-PSPs) on hyperglycemia and hyperuricemia induced by a high-fructose/high-fat diet. The body weight, organ index, serum biochemical indexes, and liver antioxidant indexes of mice were measured, and the kidneys were observed in pathological sections. The relative expression levels of messenger RNAs (mRNAs) of fructose metabolism pathway enzymes in kidney were detected by fluorescent real-time quantitative polymerase chain (qPCR) reaction technique, and the expression of renal transporter protein and inflammatory factor pathway protein was determined by immunohistochemistry (IHC) technique. Results showed that diacylated AF-PSPs alleviated hyperuricemia in mice, and that this effect might be related to the regulation of liver XO activity, lipid accumulation, and relevant renal transporters. Diacylated AF-PSPs reduced body weight and relieved lipid metabolism disorder, liver lipid accumulation, and liver oxidative stress, thereby enhancing insulin utilization and sensitivity, lowering blood sugar, and reducing hyperglycemia in mice. Also, diacylated AF-PSPs restored mRNA levels related to renal fructose metabolism, and reduced kidney injury and inflammation. This study provided experimental evidence for the mechanisms of dual regulation of blood glucose and uric acid (UA) by diacylated AF-PSPs and their utilization as functional foods in the management of metabolic syndrome.

ABSTRACT

The flavonoids in Panax notoginseng were qualitatively analyzed by ultra-high performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS), and the content of three main flavonoids in P. notoginseng of different specifications and grades collected from different habitats was determined by HPLC-DAD. Flavonoids and anthocyanins were analyzed by UPLC-Q-TOF-MS/MS in the positive and negative ion modes, respectively. Twelve flavonoid glycosides and one anthocyanin glycoside in P. notoginseng were identified, but no flavonoid aglycones were detected. Among them, 12 compounds were identified in the underground part of P. notoginseng for the first time and eight compounds were first reported in this plant. Moreover, six and four compounds were identified in the Panax genus and the Araliaceae family for the first time, respectively. A method for simultaneous determination of three flavonoids in P. notoginseng was established by HPLC-DAD. The content of flavonoids in 721 P. notoginseng samples of 124 specifications and grades collected from 20 different habitats was simultaneously determined. Among three flavonoids determined, the content of quercetin-3-O-(2″-β-D-xylosyl)-β-D-galactoside was the highest with the average content in the tested samples of 161.0 μg·g~(-1). The content of compounds quercetin-3-O-hexosyl-hexoside and kaempferol-3-O-pentosyl-hexoside was relatively low, with the average content of 18.5 μg·g~(-1)(calculated as quercetin-3-O-sophoroside) and 49.4 μg·g~(-1)(calculated as kaempferol-3-O-sangbu diglycoside). There were significant differences in flavonoids content of samples from different production area. The content of flavonoids in spring P. notoginseng was significantly lower than that in winter P. notoginseng when the other influencing factors such as production areas, germplasm resources, and cultivation conditions were fixed. As for P. notoginseng of different specifications, the flavonoid content in the part connecting the taproot and the aboveground stem was significantly higher than that in other parts. The results of large-scale data showed that the flavonoid content gradually increased with the increase in the number of heads. There were significant differences between the flavonoid content in most specifications and grades, especially the 20-head P. notoginseng and countless head P. notoginseng, whose content was significantly lower and significantly higher than that of other specifications and grades, respectively. This study provides a scientific basis for the study of the effective components and quality control of P. notoginseng from the perspective of flavonoids.

ABSTRACT

Abstract The hydroelectrolytic beverages segment has been expading its market and introducing new flavors in order to meet the demand for new products. However, experimental studies find concerns about the chemical compositions of these drinks. The aim of this study was to develop a drink without synthetic coloring or flavoring, with functional attributes based on the bacaba (Oenocarpus bacaba Mart.) peel extract. Two hydroelectrolytic drinks were developed, one hypotonic and the other isotonic, containing 0.5 and 1.0% of bacaba peel extract. Physicochemical characterization, determination of total phenolic compounds, anthocyanins, and antioxidant capacity were perfomed, in addition to color evaluation, as well as sensory analysis by means of preference tests. The developed formulations showed potential antioxidant activity and natural red coloring due to the phenolic compounds and anthocyanins present in the beverages. The sensory evaluation indicated positive acceptance by the tasters regarding the addition of the bacaba peel extract to the beverage formulations. The developed formulations demonstrated that the use of the bacaba peel is a viable option for the production of sports drinks, acting as a natural dye and offering health benefits due to its bioactive compounds.

We investigated the effects of the juçara fruit (Euterpe edulis Martius) pulp and lyophilized extract on the expression of cytoprotective genes nuclear factor erythroid 2 (NF-E2)-related factor 2 (NRF2), kelch-like ECH-associated protein 1 (KEAP1), superoxide dismutase (SOD1), and glutathione peroxidase (GPX2) in human colorectal cancer cell lines (HT-29 and Caco-2). Cells were cultured for 24 h in Dulbecco's Modified Eagle's Medium containing juçara fruit pulp (5, 10, or 50 mg/mL) or lyophilized extract (0.05, 0.1, or 0.5 mg/mL), and gene expression was quantified using real-time quantitative reverse transcription polymerase chain reaction. All studied genes showed significant variation in gene expression among different concentrations of pulp or lyophilized extract. Overall, the expression of the selected genes decreased in both cell lines following exposure to the pulp or lyophilized extract in a dose-dependent manner for most of the concentrations studied. In summary, our study showed that the compounds in juçara fruit inhibited the expression of cytoprotective genes associated with the antioxidant response and that, although not cytotoxic at the concentrations studied, they could potentially block the activation of the NRF2/KEAP1 pathway.

Anthocyanins belong to flavonoids, which have the characteristics of antioxidant properties, and have the functions of protecting liver, improving memory, improving immunity, and protecting nervous system, digestive system, urinary system, cardiovascular system, etc. In this paper, the research status of arsenic-induced health hazards and the protective mechanism of anthocyanins on health hazards in recent years is summarized, in order to provide scientific basis for prevention and treatment of chronic arsenic poisoning injury.

ABSTRACT

Anthocyanins are widely distributed water-soluble pigments that not only give the fruit colorful appearances, but also are important sources of natural edible pigments. In recent years, the interest on anthocyanins of solanaceous vegetables is increasing. This paper summarized the structure of anthocyanins and its biosynthetic pathway, the structural genes and regulatory genes involved in the biosynthesis of anthocyanins in solanaceous vegetables, as well as the environmental factors affecting the biosynthesis. This review may help clarify the synthesis and regulation mechanism of anthocyanins in solanaceous vegetables and make better use of anthocyanins for quality breeding of fruit colors.

Resumen La investigación tuvo como objetivo definir las mejores condiciones de extracción asistida por ultrasonido de los cálices de H. sabdariffa L. y la obtención de polvos microencapsulados, mediante secado por aspersión. Los extractos fueron analizados, considerando como variables: disolvente (agua y agua/etanol) y la relación temperatura/tiempo de extracción (25 °C/60 min y 60 °C/30 min). Para el secado se evaluaron las variables temperatura de entrada (150 °C; 190 °C) y la mezcla de encapsulantes goma arábiga (G) y maltodextrina (MD) (G40/ MD60; G60/MD40). Los parámetros utilizados para el análisis fueron: rendimiento, pH, °Bx, composición química (fenoles y antocianinas totales, CLAE-EM) y capacidad antioxidante (DPPH). La mejor condición para la extracción de polifenoles resultó ser con agua:etanol (80:20), a 60 °C y durante 30 min. Se identificó la presencia de ácidos fenólicos, glicósidos de flavonoles y las antocianinas (delfinidina-3-sambubiósido y cianidina-3-sambubiósido), como las señales de mayor intensidad. Con el secado por atomización a 150°Cy con G60/MD40, se logró el mayor contenido de fenoles totales y antocianinas, sin embargo, la capacidad antioxidante se favoreció a 150 °C y con G40/MD60. Las micropartículas obtenidas podrían valorarse como materia prima para la elaboración de fitofármacos o alimentos funcionales, considerando su fácil manipulación, posible estabilidad y su valor antioxidante.


Abstract The objective of the research was to define the best conditions for ultrasound-assisted extraction of H. sabdariffa L. calyces, and to obtain microencapsulated powders, by spray drying. The extracts were analyzed, considering as variables: extracting solvent (water and water/ethanol) and the temperature /extraction time ratio (25 °C/ 60 min and 60 °C/30 min). Inlet air temperature (150 °C; 190 °C) and the mixture of gum arabic (G) and maltodextrin (MD) as encapsulating agents (G40/MD60; G60/MD40) were the variables studied. The parameters used for the analysis were: yield, pH, °Bx, chemical composition (phenols and total anthocyanins, HPLC-MS), and antioxidant capacity (DPPH). The best polyphenols extraction conditions were water:ethanol (80:20), at 60 °C for 30 min. The presence of phenolic acids, flavonol glycosides, and anthocyanins (delphinidin-3-sambubioside and cyanidin-3-sambubioside) were identified as the signals of highest intensity. Inlet air temperature at 150 °C and G60/MD40 allowed the highest total phenols and anthocyanins content. However, the antioxidant capacity was better at 150 °C and G40/MD60. The microparticle obtained could be used as an ingredient for the preparation of phytopharmaceuticals or functional foods, considering their easy handling, and antioxidant capacity.


Resumo O objetivo da pesquisa foi definir as melhores condições para a extração assistida por ultrassom de H. sabdariffa L. calyces e obter pós microencapsulados, por meio de secagem por pulverização. Os extratos foram analisados considerando-se variáveis: menstruação (água e água/etanol) e a razão temperatura/tempo de extração (25 °C/60 min e 60°C/30 min). Para a secagem, os variais foram avaliados: temperatura de entrada (150 °C; 190 °C) e mistura dos encapsulantes goma arábica (G) e maltodextrina (MD) (G40/ MD60; G60/MD40). Os parâmetros utilizados para a análise foram: rendimento, pH, °Bx, composição química (fenóis e antocianinas totais, HPLC-MS) e capacidade antioxidante (DPPH). A melhor condição de extração acabou com a água: etanol (80:20), a 60 °C e por 30 min. A presença de ácidos fenólicos, flavonol glicosídeos e antocianinas (delfinidin-3-sambubiosídeo e cianidin-3-sambubiosídeo) foram identificados como sinais de maior intensidade. Com a secagem por pulverização a 150°Ce com G60/MD40, foi atingido o maior teor de fenóis e antocianinas totais, no entanto, a capacidade antioxidante foi favorecida a 150 °C e com G40/MD60. As microcápsulas obtidas podem ser utilizadas como matéria-prima na preparação de fitofarmacêuticos ou alimentos funcionais, considerando seu fácil manuseio, possível estabilidade e seu valor antioxidante.

ABSTRACT: "Araçá" has been reported with different biological activities such as antioxidant, antiproliferative and antimicrobial as well as inhibitors of digestive enzymes. The digestive pancreatic lipase enzyme plays a fundamental role in lipid metabolism, and its inhibition has been studied as a target for obesity treatment. This study quantified the bioactive compounds present in different parts of "araçá" fruit and evaluated their antioxidant activity and lipase inhibition properties. Three samples were analyzed for total anthocyanins, total phenolic content, antioxidant activity and pancreatic lipase inhibition. Anthocyanins were reported only in pulp-peel of red "araçá" sample. Phenolic compounds concentration was higher in pulp-peel than in seeds for all samples. The antioxidant activity followed the same trend. A positive correlation was observed between total phenolic content and both antioxidant activity and lipase inhibition. Lipase inhibition activity was higher for pulp-peel compared to the seeds. Overall, the results showed that "araçá" fruit extracts could be beneficial for the treatment of obesity.


RESUMO: A fruta araçá tem sido destacada por suas diferentes atividades biológicas como antioxidante, antiproliferativa e antimicrobiana, além de inibidores de enzimas digestivas. A enzima digestiva lipase tem papel fundamental no metabolismo lipídico, e sua inibição tem sido estudada como alvo para o tratamento da obesidade. Este estudo teve como objetivo quantificar os compostos bioativos presentes em diferentes partes do fruto do araçá e avaliar sua atividade antioxidante e propriedades de inibição da lipase. Três amostras foram analisadas quanto a antocianinas totais, compostos fenólicos totais, atividade antioxidante e inibição da lipase pancreática. Antocianinas foram encontradas apenas na polpa-casca da amostra de araçá vermelho. A concentração de compostos fenólicos foi maior na polpa-casca do que nas sementes para todas as amostras. A atividade antioxidante seguiu a mesma tendência. Os compostos fenólicos totais, a atividade antioxidante e a inibição da lipase apresentaram uma correlação positiva. A atividade de inibição da lipase foi ligeiramente superior para a polpa-casca em comparação com as sementes. No geral, os resultados revelaram que os extratos da fruta do araçá podem ser benéficos para o tratamento da obesidade.

Abstract The effect of irradiation through UV-C on the content of phenolic compounds, antioxidant activity, color, and texture profile was evaluated in Bordô variety grapes. Grapes were subjected to stress by irradiation at time of 0.5, 1.0, 4.0, 10 and 30 min that correspond at doses of 0.1, 0.3, 1.0, 2.4, and 7.2 kJ m-2, respectively. The total phenolic and anthocyanins content, antioxidant activity, color and texture of the grapes were evaluated after storage at 22 ºC for 24 and 48 h. In general, samples irradiated did not show an increase in total phenolic content, anthocyanin compounds and antioxidant activity. Hue and Chroma values indicated that the color of grapes did not change when irradiated. Texture profile analysis suggested that UV-C treatment delayed grapes softening.

ABSTRACT: Biofortification of food crops is implemented through the application of mineral fertilizers, to improve the levels of essential mineral elements for human nutrition. Magnesium is a key macronutrient in crop production and quality; however, worldwide, it is the most limiting macronutrient in agriculture. Magnesium plays an important role in manipulating physiological and biochemical processes in plants. Therefore, the objective of this study was to evaluate the efficacy of biofortification with magnesium chloride and sulfate on the accumulation of bioactive compounds and antioxidant capacity in snap bean cv. Strike. Two sources of Magnesium were applied via edaphic route: Magnesium chloride and magnesium sulfate at doses of 0, 50, 100 and 200 ppm during 2018 in Mexico. Accumulation of bioactive compounds (total phenols, total flavonoids and total anthocyanins) and antioxidant capacity in snap bean fruits were evaluated. Results obtained indicate that the concentration of total flavonoids, total anthocyanins and antioxidant capacity in the edible parts of snap beans were higher in the MgSO4 treatment than in the MgCl2 treatments, exceeding 30, 59 and 6% respectively. This is one of the first studies on the edaphic agronomic biofortification of Mg+ and its effect on nutraceutical quality in snap bean. An interesting result is that MgSO4 produced high concentrations of anthocyanins in edible snap beans. These results can be applied as a new strategy to reduce malnutrition and improve the health of the population in poor urban and rural communities in developing countries.


RESUMO: A biofortificação de culturas alimentares é implementada através da aplicação de fertilizantes minerais, para melhorar os níveis de elementos minerais essenciais para a nutrição humana. O magnésio é um macronutriente essencial na produção e qualidade das culturas, no entanto, em todo o mundo, é o macronutriente mais limitante da agricultura. O magnésio desempenha um papel importante na manipulação de processos fisiológicos e bioquímicos nas plantas. Portanto, o objetivo deste estudo foi avaliar a eficácia da biofortificação com cloreto e sulfato de magnésio no acúmulo de compostos bioativos e capacidade antioxidante em feijões verdes cv. Strike. Duas fontes de magnésio foram aplicadas por via edáfica: cloreto de magnésio e sulfato de magnésio nas doses de 0, 50, 100 e 200 ppm durante o ano de 2018 no México. Foram avaliados o acúmulo de compostos bioativos (fenóis totais, flavonóides totais e antocianinas totais) e a capacidade antioxidante em frutos de feijão-vagem. Os resultados obtidos indicam que a concentração de flavonoides totais, antocianinas totais e capacidade antioxidante nas partes comestíveis do feijão-vagem foram maiores no tratamento com MgSO4 do que nos tratamentos com MgCl2, ultrapassando 30, 59 e 6% respectivamente. Este é um dos primeiros estudos sobre a biofortificação agronômica edáfica do Mg+ e seu efeito na qualidade nutracêutica do feijão-vagem. Um resultado interessante é que o MgSO4 produziu altas concentrações de antocianinas no feijão-vagem comestível. Esses resultados podem ser aplicados como uma nova estratégia para reduzir a desnutrição e melhorar a saúde da população em comunidades urbanas e rurais pobres em países em desenvolvimento.

ABSTRACT

miRNAs are important regulators of plant gene expression. There are few studies on the regulation of miRNAs in Lonicera edulis. We used high-throughput sequencing technology to analyse miRNAs in L. edulis, aiming to identify miRNAs and elucidate their function in L. edulis. In the present study, we employed the high-throughput sequencing technology to profile miRNAs in L. edulis. A total of 51,819,072 small RNA tags with sizes ranging from 18 to 30 nt were obtained, indicating that L. edulis have a large and diverse small RNA population. Bioinformatic analysis identified 507 mature miRNAs, and 16 predicted novel miRNAs that are likely to be unique to L. edulis. Three miRNAs related to anthocyanin biosynthesis were locked by gene ontology (GO) analysis and target gene analysis. The selected three miRNAs are relatively high in the expression of L. edulis. Some of the previous studies have studied these types of miRNAs involved in the anthocyanin metabolism pathway in fruits. Among them, expression profiles of three conserved miRNAs were validated by stem loop qRT-PCR. Further, the potential target genes of conserved and novel miRNAs were predicted and subjected to GO annotation. Enrichment analysis of the GO-represented biological processes and molecular functions revealed that these target genes were potentially involved in a wide range of metabolic pathways and developmental processes. In particular, different families of miRNAs can directly or indirectly regulate anthocyanin biosynthesis. In recent years, the research on miRNAs has become more and more clear, but the research on miRNAs involved in the regulation of anthocyanin synthesis of L. edulis is still lagging. This study provides a useful resource for further elucidation of the functional roles of miRNAs during fruit development and ripening

ABSTRACT The aim of this study was to develop and evaluate the antioxidant characteristics and sensory acceptance of Greek yogurt with added whole sorghum flours (WSF) from genotypes BRS305 (tannin) and BR501 (tannin-free). Five formulations were elaborated: 1) Control (without WSF); 2) 2% BR501; 3) 4% BR501; 4) 2% BRS305 and 5) 4% BRS305 and evaluated for antioxidant capacity, total anthocyanins, total phenols, condensed tannins, sensory acceptance and purchase intention. The 4% BRS305 formulation presented higher antioxidant properties, but lower sensory acceptance. No significant difference was observed for: color, flavor and overall acceptability between the control and 2% BR501; nor for texture and overall acceptability between the control and 4% BR501. Principal component analysis explained that all variables associated with antioxidant properties were positively correlated with the first major component (PC1: 82.7%). The 2% and 4% BR501 formulations were more highly correlated with PC1 (92.7%), as well as the control in relation to sensorial characteristics. The addition of WSF BRS305 improved the antioxidant properties of yogurts and the addition of WSF BR501 did not interfere with the sensory acceptance of the formulations. The similarity of the sensory acceptance of the yogurt containing WSF from the BR 501 genotype to the control, opens perspectives for the insertion of sorghum in human food, using dairy products as suitable matrices, adding potential functionality to this type of product.


RESUMEN El trabajo tuvo como propósito desarrollar y evaluar las características antioxidantes y la aceptación sensorial de yogurt griego con adición de harinas integrales de sorgo (HIS) de los genotipos BRS305 (con taninos) y BR501 (sin taninos). Cinco formulaciones fueron elaboradas: 1) Control (sin HIS); 2) 2% BR501; 3) 4% BR501; 4) 2% BRS305 y 5) 4% BRS305. Las formulaciones fueron evaluadas en función de su actividad antioxidante, antocianinas totales, fenólicos totales, taninos condensados, aceptación sensorial e intención de compra. La formulación 4% BRS305 presentó más propiedades antioxidantes, pero menor aceptación sensorial que las otras formulaciones. No se observó diferencia significativa entre el control y el 2% BR501 para el color, sabor e impresión global, ni tampoco para textura e impresión global en relación al 4% BR501. El análisis de componente principal ha explicado que todas las variables asociadas a las propiedades antioxidantes han sido positivamente correlacionadas con el primer componente principal (PC1: 82,7%). Las formulaciones 2% y 4% BR501 se han correlacionado más con el PC1 (92,7%), así como el control con las características sensoriales. La adición de HIS BRS305 ha mejorado las propiedades antioxidantes de los yogurts y la adición de HIS BR501 no ha interferido en la aceptación sensorial de las formulaciones. La similitud de la aceptación sensorial del yogurt que contiene el HIS del genotipo BR501 con el control, abre perspectivas para la inserción del sorgo en la alimentación humana, utilizando productos lácteos como matrices adecuadas, agregando funcionalidad potencial a este tipo de producto.

ABSTRACT

Syzygium cumini or Eugenia Jambolana Lam is a traditional medicinal plant very native to the Indian and Asian sub-continent. It is commonly known as black plum or jamun. Various plants parts of the tree are known for ethno-medicinal uses, and in particular, the fruits of Jamun tree are well known for medicinal uses and preparation of health drinks. Studies have shown that the berries contain carbohydrates, minerals and the pharmacologically active phytochemicals. The active phytochemical includes flavonoids, terpenes, and anthocyanins. Ayurvedic and Indian Folk Medicine have already mentioned the use of jamun for diabetic treatment even before the advent of insulin. The bark, fruit pulp, seeds and the leaves are evaluated in the crude form as well as extracts in various diabetic models. Further, bioactive phytochemicals are extracted using in vitro assays, some of them such as vitalboside-A, mycominose are also characterized with spectroscopic analysis. This review article is intended to compile Syzygium cumini’s therapeutic potential with special emphasis on the phytochemical constituents and its pharmacological activities related to diabetic intervention through recent studies conducted in last one decade. It also addresses the clinical and toxicological studies.

Background:Vaccinium meridionale produces fruits with a high content of anthocyanins and polyphenols with great antioxidant capacity. Objective: produce vinegar from V. meridionale alcoholic beverages and to determine the content of bioactive antioxidant compounds, antioxidant capacity, and cytotoxic activity on colorectal cancer cells. Methods: Wine and vinegar samples were obtained by fermentation of V. meridionale berries juice, using three extractive processes: mechanical maceration (MAC), preheating to 80°C (CAL) and a combination of both (MIX). During acetic fermentation, titratable acidity and acetic acid content were evaluated. Fermentation progress was recorded and compared by measuring the antioxidant potential by DPPH, FRAP, and ORAC. Polyphenols, anthocyanins and hydroxycinnamic acids were quantified. Finally, the antiproliferative activity of vinegar was evaluated in SW480 colon cancer cells. Results: In acetic fermentation, yield and productivity were independent of extraction, indicating that they do not affect the biotransformation of alcohol into vinegar. The alcoholic beverages showed the highest antioxidant activity; after acetic fermentation, a decrease in antioxidant potential was observed in all three extractive processes evaluated. The different vinegar obtained by CAL and MIX, showed the highest values of antioxidant activity, polyphenols, and anthocyanins. The inhibition of the antiproliferative activity of vinegar was dose-dependent and showed an IC50 of 536 µg/mL. Conclusions: The vinegar prepared from V. meridionale berries presented an outstanding antioxidant and antiproliferative activity. The reason is the contents of bioactive compounds and their antioxidant power, which may contribute to chemoprevention in secondary cancer prevention.


Antecedentes: Vaccinium meridionale produce frutos con alto contenido en antocianinas y polifenoles con elevada capacidad antioxidante. Objetivo: Producir vinagre a partir de bebidas alcohólicas de V. meridionale y determinar el contenido de compuestos bioactivos antioxidantes, capacidad antioxidante y actividad citotóxica sobre células de cáncer colorrectal. Métodos: Muestras de Vino y vinagre fueron obtenidas mediante fermentación del jugo de V. meridionale, usando tres procesos extractivos: maceración mecánica (MAC), precalentamiento a 80 °C (CAL) y una combinación de ambos (MIX). En la fermentación acética fueron evaluadas la acidez titulable y el contenido de ácido acético. Se registró y comparó la evolución de la fermentación midiendo el potencial antioxidante por DPPH, FRAP y ORAC. Se cuantificaron los polifenoles, antocianinas y ácidos hidroxicinámicos. Finalmente, la actividad antiproliferativa del vinagre fue evaluada en células de cáncer de colon SW480. Resultados: En la fermentación acética, el rendimiento y la productividad fueron independientes de la extracción indicando que no afectan la biotransformación del alcohol en vinagre. Las bebidas alcohólicas mostraron la mayor actividad antioxidante, después de la fermentación acética se observó una disminución del potencial antioxidante en los tres procesos extractivos evaluados. Los vinagres obtenidos por CAL y MIX, presentaron los mayores valores de actividad antioxidante, polifenoles y antocianinas. La inhibición de la actividad antiproliferativa del vinagre fue dosis dependiente y mostró un IC50 de 536 µg/mL. Conclusiones: El vinagre preparado a partir de bayas de V. meridionale presentó una excelente actividad antioxidante y antiproliferativa debido al contenido de compuestos bioactivos y poder antioxidante que puede contribuir a la quimioprevención en la prevención secundaria del cáncer.

Contrast-induced nephropathy (NIC) is directly related to increased morbidity and mortality, and its treatment and prevention might be achieved by the administration of antioxidant products. The juçara palmetto (Euterpe edulis Martius) has fruits rich in phenolic compounds, which are known for their antioxidant activity. This work aimed to evaluate the nephroprotective activity of E. edulis pulp in the NIC animal model. The collected fruits were pulped, their contents of polyphenols and anthocyanins were quantified, and their antioxidant activity were evaluated. The nephroprotective effects were determined based on iodine contrast induction and evaluated by biochemical and histological analyses. The results showed that E. edulis pulp was rich in polyphenols (811 ± 16.7 mg EAG/g) and anthocyanins (181.25 mg/100 g) and had very strong antioxidant activity, as demonstrated by the DPPH (2,2-diphenyl-1-picryl-hydrazyl) method, which revealed an antioxidant activity index (AAI) of 3.4, and the 2,29-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) method, which revealed an IC50 of 0.59 ± 0.03 mg/mL. In the in vivo experiments, E. edulis pulp tended to provide renal protection and reduce renal dysfunction and tubular morphological lesions in mice after the induction of NIC, and these effects were obtained through the antioxidant activities of the polyphenols in the pulp.

Little is known about the molecular mechanism of currant anthocyanin synthesis. We investigated the effect of dfr, a key gene for anthocyanin synthesis in currant, on anthocyanins of different color currant. Black currant (Ribes nigrum L.), red currant (Ribes rubrum L.) and white currant (Ribes albrum L.) were used as test materials to determine the anthocyanin content at different stages of fruit development. Three full-length cDNA sequences of dfr gene were cloned by RACE (Rapid amplification of cDNA ends), and named as Rndfr, Rrdfr and Radfr. Phylogenetic analysis shows that Rndfr, Rrdfr and Radfr had high homology in evolution. The determination of anthocyanin content in different stages of fruit development shows that the content of anthocyanin in black currant and red currant was higher and gradually increased with the ripening of the fruit. While the content of anthocyanin in white currant was extremely low, and almost no anthocyanin was detected. Quantitative RT-PCR analysis shows that the expression level of dfr in black currant was higher than red currant and white currant in each period of fruit development. As the diameter of the fruit increased and the color of the peel deepened, the expression level of dfr in the black currant showed an increasing trend. In the red currant, the expression level gradually increased until the period of 75% fruit color, then the Rrdfr decreased rapidly. In white currant, the overall trend showed a downward trend, and its expression level was the lowest. All the results suggest that dfr gene plays a role in the process of fruit color.

Background: Foods including probiotics are considered "functional foods." As an alternative to dairy products, we investigated the behavior of Lactobacillus casei when exposed to low-pH fruit juice. Juices of fruits such as pineapple, raspberry, and orange were assessed. Free and microencapsulated forms of L. casei were compared, and the viability of the probiotic was evaluated under storage at 4°C for 28 d. Microbiological analyses were carried out to ensure a safe and healthy product for consumers who look for foods with probiotics from sources other than dairy. Results: Low pH affected L. casei survival during storage depending on the type of fruit juice. In the case of pineapple juice, some microcapsules were broken, but microcapsules recovered at the end of the storage period had 100% viability (2.3 × 107 CFU/g spheres). In the case of orange juice, more than 91% viability (5.5 × 106 CFU/g spheres) was found. In raspberry juice, viability decreased rapidly, disappearing at the end of the storage period, which was caused by the absorption of high concentrations of anthocyanin inside microcapsules more than low pH. Conclusion: Low pH affected the survival of L. casei under refrigeration; even when they were microencapsulated, acidic conditions impacted their viability. Although pH affects viability, its value is very sensitive and will depend on the type of fruit juice and its composition. Some fruit juices contain compounds used as substrates for Lactobacillus and other compounds with antimicrobial effects.

Phytochemical-enriched edible greens, sweet potato leaves (Ipomoea batatas L.), have become popular due to potential health benefits. However, the phytochemical contents in sweet potato leaves and their subsequent change over harvest stages and growth condition are mostly unknown. In this study, the anthocyanin profile and content in leaves of four sweet potato cultivars, i.e., white-skinned and white-fleshed Bonita, red-skinned and orange-fleshed Beauregard, red-skinned and white-fleshed Murasaki and purple-skinned and purple-fleshed P40, were evaluated. Fourteen anthocyanins were isolated and identified by HPLC-MSI/MS. The most abundant was cyanidin 3-caffeoyl-p-hydroxybenzoyl sophoroside-5-glucoside, which comprised up to 20% of the total anthocyanins. Of the young leaves (1st and 2nd slip cuttings), Bonita contained the highest anthocyanin content followed by P40. Of the mature leaves (vine stage), Beauregard had the greatest anthocyanin (592.5 ± 86.4 mg/kg DW) and total phenolic (52.2 ± 3 mg GAE/g DW). It should be noted that the lowest anthocyanin and total phenolic content of shoots were found in P40, while tubers of P40 contain the highest content of each. Furthermore, the increase in leaf anthocyanin content over the growth stages that was observed in three of the cultivars but not in P40. No significant difference of anthocyanin content was found in Beauregard leaves grown in the high tunnels when compared with that in the open field. This study demonstrated for the first time that anthocyanin levels were significantly changed in response to various growth stages but not high tunnel condition, indicating that the effect of anthocyanin biosynthesis in sweet potato leaves is highly variable and genotype specific.
